Isopullulanase

I hydrolyze α-1,4-glucoside bond of the many sugar pullulan, and the isopullulanase (Isopullulanase, IPU; EC 3.2.1.57) is an enzyme producing trisaccharides isopanose. The systematic name is pullulan 4-glucanohydrolase (isopanose-forming). GH49 family.

Summary

I was found than black Aspergillus oryzae Aspergillus niger ATCC9642 stock in 1971 by Yoshiyuki Sakano and others [1].

Glcα-(1 → 6 that it is the partial structure of the starch) -Glcα-(1 → 4) -I break α-1,4-glucoside bond of the oligosaccharide with structure called Glc, but do not act on starch itself. The enzyme breaking down pullulan has pullulanase (EC 3.2.1.41) and neo-pullulanase (EC 3.2.1.135) other than IPU, but all are classified in glucide hydrolase family 13 (GH13) in CAZy[2]. IPU is classified in GH49 for it, and other pullulan degrading enzymes and homogeny in the primary structure are not accepted at all. An enzyme belonging to GH49 is the enzyme that dextranase (EC 3.2.1.11) hydrolyzes many sugar dextran consisting of α-1,6-glucoside combination except IPU, but IPU has the characteristic that I cannot break down dextran into.

In 2008, a tertiary structure became clear by an X-ray crystallographic analysis [3]. IPU is comprised of an N-terminal side domain consisting of the backward movement beta sheet and a C-terminal side domain to take β ヘリックス structure that three β strands form 1 coil, and it affects ten.
